چکیده

The large surface area and the short diffusion distance from capillaries of the blood brain barrier (BBB) to the neurons facilitate the drugs and nutrients access to the brain. Penetration of chemicals to the BBB occurs using a combination of intra and intercellular passages. Tight junctions regulate the intracellular passage of molecules according to their physicochemical properties (e.g. lipophilicity, ionisation and polarity), where inter cellular penetration is regulated by influx and efflux transporters, endocytosis and passive diffusion. Poor pharmacokinetic properties (absorption, distribution, metabolism and excretion) and toxicity are responsible for most of the failures in drug discovery projects. This problem is more evident for CNS drugs because of the restrict barrier function of blood brain barrier. The CNS drug discovery attracted more attentions since the diseases pattern has been changed during recent decades and aging disorders are one of the major health problems. Drug exposure is controlled by plasma pharmacokinetic properties of drug which are different from brain pharmacokinetic and can be studied using common pharmacokinetic studies, where BBB permeability depends on physicochemical properties of drug compound and physiologic function of the BBB (physical barrier, transport, metabolic, ...) and need special study techniques. In this chapter, fundamentals of BBB, permeation mechanisms, penetration measurement methods and penetration prediction methods are discussed.

In silico prediction of blood-brain barrier permeation.

This review examines the progress that is being made towards the in silico prediction of brain permeation. Following a brief introduction to the blood-brain barrier, the datasets currently available for in silico modeling are discussed. Recent developments in in silico models of brain permeation are summarized in the context of the current state of the art in prediction accuracy. An analysis of...

Preparation of Silica Nanoparticles Loaded with Nootropics and Their In Vivo Permeation through Blood-Brain Barrier

The blood-brain barrier prevents the passage of many drugs that target the central nervous system. This paper presents the preparation and characterization of silica-based nanocarriers loaded with piracetam, pentoxifylline, and pyridoxine (drugs from the class of nootropics), which are designed to enhance the permeation of the drugs from the circulatory system through the blood-brain barrier. T...

Deoxycholic Acid as a Modifier of the Permeation of Gliclazide through the Blood Brain Barrier of a Rat

Major problem for diabetic patients represents damage of blood vessels and the oxidative stress of the brain cells due to increased concentration of free radicals and poor nutrition of brain cells. Gliclazide has antioxidative properties and poor blood brain barrier (BBB) penetration. Bile acids are known for their hypoglycemic effect and as promoters of drug penetration across biological membr...

Synthesis and analgesic activity evaluation of some agmatine derivatives.

A series of N,N'-disubstituted-2-nitroethene-1,1-diamine and N,N'-disubstituted- N''-cyanoguanidine derivatives were prepared and evaluated for in vivo analgesic activity. The blood brain barrier (BBB) VolSurf model was used to predict the BBB permeation profiles of our synthesized compounds. Some compounds show both remarkable analgesic activity and good BBB permeation profiles, and these comp...

Blood-Brain Barrier Permeation of Glutathione-Coated Nanoparticle

The blood-brain barrier (BBB) is a physical interface in the central nervous system (CNS) that exists between the blood and neural tissue. The phenotype of the BBB is primarily the result of the interactions between three main cell types: Endothelial cells, astrocytes, and pericytes [1]. The interactions of the endothelial cells and astrocytes seems to be of utmost importance in maintaining the...
